Can Peptides Reverse Ageing? The Anti-Ageing Peptide Protocol

By Samantha Miller, Clinical Skin Therapist & Founder of The Naked Chemist | Updated: August 2026

Anti-ageing skincare is a minefield.

Miracle this, breakthrough that, years off your face by Friday.

It’s exhausting, and most of it is just noise.

But every so often an ingredient earns the hype — and peptides are that rare thing.

Here’s the catch: their names are complex.

So I’ve put together this anti-ageing peptide protocol.

What each one does, and how to spot them on a label.

Deep breath. Let’s decode them together.

Direct Answer

An anti ageing peptide protocol is a consistent routine built around peptides — short chains of amino acids that signal your skin to repair itself. Different peptides do different jobs: some boost collagen and elastin, some relax expression lines, and some calm inflammation or even reduce the appearance of pigmentation. Used daily and consistently, they help rebuild the skin’s foundation, improving firmness, texture and resilience over 8 to 12 weeks.“This guide covers peptides you apply to your skin, not injectables or supplements”.

Key Takeaways

  • Peptides are short chains of amino acids that act as messengers, signalling your skin to repair, rebuild and behave more youthfully.
  • There are five main types — carrier, signalling, enzyme-inhibiting, antimicrobial and neurotransmitter-inhibiting — each with a distinct job.
  • The best-researched include copper tripeptide (GHK-Cu) for repair, Matrixyl for fine lines, and Argireline for expression lines.
  • Peptides complement, rather than replace, retinol and pair beautifully with ceramides and antioxidants.
  • Consistency is everything — a real peptide protocol means daily use for 8 to 12 weeks, not occasional applications.

What Peptides Actually Do

Peptides work as tiny messengers. When applied to the skin, they signal your cells to produce specific tissue types that promote healing and repair — in effect, they gently trick your skin into thinking it needs to build more protein.

Here at NC HQ, I liken them to a light switch: they signal specific skin functions to turn on and off. Through active amino acid sequences, they can induce or inhibit the formation of specific proteins, which is why the right formulation matters so much. When these short chains are produced in a specific way, they help rebuild the proteins that give your skin its foundational support: elastin, keratin and collagen. All three decline with age, which is where a good peptide protocol comes into play.

The Five Types of Peptide

1. Signal peptides

These send your skin a message: make more collagen and elastin. Think of them as a nudge that says “time to rebuild.” They’re the most widely used peptides in anti-ageing skincare.

Example: palmitoyl pentapeptide-4 (Matrixyl).

2. Carrier peptides

These act like a delivery service. They carry tiny amounts of minerals, such as copper, into the skin, where your cells use them to repair and renew. Copper peptides are the best known.

Example: copper tripeptide-1 (GHK-Cu).

3. Enzyme-inhibiting peptides

Your skin has enzymes that break down collagen, and others that drive pigmentation. These peptides slow them down, so you keep more of the collagen you already have, and your skin tone looks more even over time.

Example: soybean and rice peptides.

4. Antimicrobial peptides

Your skin makes these naturally as part of its first line of defence. They help keep unwanted bacteria and fungi in check, support a healthy barrier and help calm redness. That makes them especially useful for damaged, reactive, or recovering skin.

Example: oligopeptide-10. Your skin also makes its own, called cathelicidins and defensins.

5. Neurotransmitter-inhibiting peptides

Often called the “Botox-like” peptides, though they’re far gentler. They quiet the signal that tells the small muscles in your face to tense, so your expression lines like crow’s feet and frown lines look softer over time.

Example: acetyl hexapeptide-8 (Argireline).

Peptides Worth Knowing by Name

A handful of peptides have strong research behind them. These are the ones worth recognising on your skincare label.

Palmitoyl pentapeptide-4 (Matrixyl)

This is the most widely used peptide in skincare. It tricks your skin into thinking collagen has broken down, prompting it to produce more—helping smooth wrinkles and plump your skin.

Acetyl hexapeptide-8 (Argireline)

This peptide eases the tension that creates expression lines, so everyday muscle movement leaves softer marks over time. It’s a favourite in eye care: in the original study, an Argireline cream reduced wrinkle depth by up to 30% after 30 days (Blanes-Mira et al., 2002, doi.org/10.1046/j.1467-2494.2002.00153.x).

Dipeptide-2 & eye-area peptides

Dipeptide-2 targets puffiness and, when combined with peptides like palmitoyl oligopeptide and tetrapeptide, works around the eyes to soften dark circles and strengthen the area’s delicate skin.

Copper tripeptide (GHK-Cu)

In a one-month study comparing collagen boosters, copper peptide increased collagen production in 70% of women, against 50% for vitamin C and 40% for retinoic acid (Abdulghani et al., 1998; reviewed in Pickart & Margolina, 2018, doi.org/10.3390/ijms19071987). It acts as both a signal and a carrier peptide, and its calming, anti-inflammatory action makes it superb for a compromised barrier, which is why we use it in our DNA Age Delay Skin Shot.

DNA Age Delay Skin Shot

Because copper tripeptide is such a multitasker—collagen, elastin, barrier repair, all at once —we use it at full concentration in our DNA Age Delay Skin Shot, alongside New Zealand Mamaku black fern. It’s the anchor of the peptide protocol: rebuilding thin, fragile skin and softening the look of fine lines, wrinkles, and broken capillaries.

Wrinkle-Relaxing Peptides

No anti-ageing peptide protocol is complete without mentioning wrinkle-relaxing peptides, which soften expression lines by easing muscle contraction, a gentler cousin of injectables. This study found peptide-based serums measurably improved expression lines and re-densified skin in women aged 35–60 after just 12 weeks of regular use.

The four you’ll see most often are:

  • Argireline: acetyl hexapeptide-8 (also labelled -3) — the best known
  • Snap-8: acetyl octapeptide-3
  • Leuphasyl: pentapeptide-18
  • Syn-Ake: dipeptide diaminobutyroyl benzylamide diacetate

These often work synergistically — Argirelox, for instance, pairs Argireline with Leuphasyl. The suppliers’ recommended concentration for this group is 5-10%, which the label should confirm.

Building Your Anti-Ageing Peptide Protocol

The single rule of a peptide protocol is consistency. Occasional use will not work effectively. Peptides change your skin’s behaviour, and that only happens with regular, continuous application as part of your daily routine.

Think of it like laying a foundation. Used faithfully, peptides rebuild your skin’s structure, making it more resilient and helping ward off premature ageing for years to come. They complement retinol rather than replace it, and layer well with ceramides, cholesterol, and fatty acids to support fragile skin. Give it 8 to 12 weeks of daily use — patience is part of the protocol.

A quick note: this protocol covers peptides you apply to your skin, not injectable or oral peptides. Those are a different conversation, best had with your healthcare professional.

My Morning Peptide Routine

In the morning, the goal is hydration and protection: plump the skin, support the barrier, and shield it from the day.

  1. Cleanse with Miracle cleansing oil
  2. DNA Age Delay Skin Shot: copper tripeptide to support collagen and calm the skin.
  3. Quench Peptide Gel on slightly damp skin to draw in and hold water.
  4. Fortify Barrier Repair Cream to seal it in.
  5. SPF every single day, rain or shine. Sun is the fastest way to undo everything your peptides are building.

My Evening Peptide Routine

At night, your skin moves into repair mode. This is when your collagen-building peptides do their best work.

  1. Cleanse to remove SPF, makeup, and the day.
  2. DNA Age Delay Skin Shot: copper tripeptide to support collagen and calm the skin.
  3. Quench to layer in more peptide support.
  4. Reset Age Defying Cream to lock everything in overnight.

Give it 8 to 12 weeks of daily use. Most people notice firmer, smoother skin first, then fine lines start to soften. Patience is part of the protocol.

Quench Peptide Gel

A good peptide protocol isn’t only about collagen. Quench pairs palmitoyl tripeptide-38, which boosts collagen and hyaluronic acid, with skin-identical humectants (HA, urea, sodium PCA) and ceramides to strengthen a fragile, reactive barrier. It’s the hydrating half of the protocol: Soothing and beautifully light, leaving your skin plump and dewy.

To Conclude. The Naked Truth

Peptides don’t just sit on your skin. They tell it what to do: make more collagen, hold on to what you’ve got, calm down, soften the lines. That’s why I rate them. “Peptide” on the front of a bottle means nothing on its own. In 30 years I’ve seen countless products with a pinch of peptide added just so they can say it, usually sitting near the bottom of the ingredient list, below the preservatives.

So read the label carefully, not the marketing. The higher a peptide sits on the ingredients list, the more of it there is. Choose well-made products, use them every day, and give them 8 to 12 weeks. That’s when you’ll see the difference.

FAQ

How do peptides actually work in skincare?

Peptides are short chains of amino acids that act as messengers, signalling your skin to perform specific tasks — from encouraging collagen to calming inflammation or boosting hydration. In simple terms, they remind the skin to behave more youthfully and repair itself more efficiently.

Can peptides replace retinol or other strong actives?

Not quite. Retinoids resurface and speed cell turnover, while peptides help rebuild the skin’s foundation from within. They’re excellent for sensitive or mature skin and pair well with ceramides and antioxidants — think complement, not replacement.

Which peptides are best for ageing or stressed skin?

Copper tripeptide (GHK-Cu) supports repair and collagen, Matrixyl helps smooth fine lines, and Argireline gently softens expression lines. Each targets a different pathway to improve firmness, texture and the look of wrinkles.

How long until I see results?

Most people notice improvements in firmness and texture within 8 to 12 weeks of consistent use. Because peptides work by influencing protein synthesis, daily application and patience are key.

Are peptides suitable for a damaged or sensitive barrier?

Yes. Many peptides — especially copper peptides and soothing signal peptides — help rebuild barrier integrity and reduce redness. They layer well with ceramides, cholesterol and fatty acids to strengthen fragile, overworked skin.

What’s your opinion on the interaction of Vitamin C and copper peptides? Concerns have been raised about the effects lost of using them together. Also, warning have been made not to use copper peptides with AHA,BHA or retinoids? Is it true? Thank you so much.

Hi Whilst we do promote the use of active anti-ageing cosmeceuticals to target expression lines and other signs of ageing, we recommend that each ingredient be used at the opposite time of day from each other, or alternated and never combine these ingredients in one formula. You have to remember they get results for a reason because they are active on your skin. Samantha
